John was the son of John Sinclair, Lord of Herdmanston and Elizabeth, daughter of John Sinclair of Stevenson.[1]

John married Catherine Sinclair, heiress of John Sinclair, 10th Lord Sinclair, they are known to have had the following issue:[1]
- Henry Sinclair, Master of Herdmanston, who also succeeded as Lord Sinclair.
