From the R334's eastern origin it leaves the N2 heading west. It runs through the northern parts of Motherwell, where it meets the southern origin of the R335, before crossing the R75 to enter Uitenhage. In Uitenhage's town centre, it meets the north-western origins of the M19 and M10. It leaves the town to the south-west, passing KwaNobuhle. It ends at the R102 to the west and just outside of Port Elizabeth, near an interchange with the N2.
